Cholyl-lys-fluorescein (CLF/cholyl-lys-F) is a bile acid (BA) analog, developed to investigate and determine in vivo liver functions.

The compound is actually a fluorescein-labeled bile acid derivative with similar properties to the naturally occurring primary bile cholic acid.. CLF is a unipolar fluorescent monohydroxy bile metabolite parallel with physiological and cholestatic properties similar to those of cholic acid, lithocholate, glycolithocholate, and more. Its molecules are excreted through bile canaliculi by the Bile Salt Efflux Pump (BSEP).

Cholyl-lys-fluorescein finds a wide range of applications.

  1. 1. It is a suitable detergent to study the inhibition of bile acid efflux transporters - a process involved in drug-induced cholestasis. Because of CLF’s cholestatic abilities, the intensity of the acid is easily measured when it’s accumulated in bile canaliculi of hepatocytes using fluorescence microscopy. In other words, it acts as a tool for visualization of bile acid transport within intrahepatic pathways.
  2. 2. CLF is useful to study mechanisms of bile salt-induced cholestasis at hepatocellular extent, as well.
  3. 3. Cholyl-lys-F has a greater biliary and hepatic excretion, compared to free fluorescein, so it’s believed that when conjugated with a bile salt, it may be an efficient approach to target the liver.
  4. 4. The fluorescent detergent undergoes minimal enterohepatic circulation. This makes CLF a proper compound to evaluate in vivo liver functions.
  5. 5. CLF is proven to show diagnostic potential to assess biliary elimination.
