Tauro-12-Oxolithocholanoic Acid is a taurine-conjugated bile acid (BA), synthesized in the liver, forming a bile salt.

The biomolecule is a derivative of 12-ketolithocholic acid (12-oxolithocholanoic acid), formed in the process of taurine conjugation by specific enzymes to create BAs with increased solubility. 12-Oxolithocholanoic acid is a keto-metabolite of lithocholic acid, a secondary BA, and an end product of the enteric bacteria.

Bile acids are amphipathic physiological detergents with steroid molecules and a range of important biological properties. They are formed in the liver as products of cholesterol metabolism, exist as anions, require a transport carrier molecule to pass through different membranes, and undergo an enterohepatic circuit. The main role of BAs is to regulate digestion, absorption, and transport of fats and fat-soluble vitamins in the intestines and liver.

They have also been reported to modulate bile flow, BAs secretion, and metabolism, as well as cholesterol, glucose, and energy homeostasis. Besides their numerous properties, essential to normal occurring biological processes, BAs have pathological effects when present in high concentrations. Accumulated BAs lead to serious cellular damage and disruption of various metabolic pathways.

In that manner, lithocholic acid and its derivatives, including 12-oxolithocholanoic acid and Tauro-12-Oxolithocholanoic acid, tend to promote neoplastic growth, when accumulated.

In addition, it is found that levels of the BA and the conjugate Tauro-12-Oxolithocholanoic Acid are elevated in patients with type 2 diabetes.
