Cholic acid 3-sulfate (disodium salt)

Cholic acid 3-sulfate (disodium salt)

3α, 7α, 12α -trihydroxy-5β-cholan-24-oic acid 3-sulfate disodium salt

Bile acids (BAs) are a large family of steroid molecules with amphipathic nature, synthesized from cholesterol in the liver. They are important for the digestion, absorption, excretion, and transport of fatty acids and other sterols in the gastrointestinal tract and liver.

On the other hand, alongside many beneficial properties, BAs may also have potent toxic effects and there are numerous mechanisms to prevent their accumulation in tissues and blood.

Cholic acid 3-sulfate (disodium salt) is a sulfated bile acid. Such sulfated bile derivatives are formed in the metabolic pathway sulfation, catalyzed by the enzyme sulfotransferase-2A1, and are far less toxic than their unsulfated bile acid counterparts. Various nuclear receptors, such as the farnesoid X receptor, pregnane X receptor, constitutive androstane receptor, and Vitamin D receptor are involved in the regulation of the sulfation reactions.

The sulfation and following synthesis of Cholic acid 3-sulfate and other sulfates are important since they transform BAs into water-soluble molecules, suppress their intestinal absorption, and promote their efficient urinary and fecal excretion. Therefore, sulfation is an essential detoxification mechanism of BAs.

So, under normal circumstances, bile acid sulfation is actually a minor metabolic pathway. However, during cholestasis (decreased bile flow and bile excretion leading to accumulation of toxic hydrophobic BAs) formation of sulfated BAs, including Cholic acid 3-sulfate, is rapidly increasing. The process takes a crucial role to maintain bile acid homeostasis under pathologic conditions.

To sum up, Cholic acid 3-sulfate and the process of sulfation itself are part of the important metabolic mechanism to neutralize toxic BAs.

In addition, Cholic acid 3-sulfate finds application in solubilizing fats and fatty derivatives to ease their biological absorption.
