Cholesterol, or rather high levels of cholesterol, can lead to heart disease and stroke which is why it gets a bed rep. But cholesterol is necessary for our bodies to build healthy cells and is even being used to deliver life-saving therapeutics. So, maybe we should rethink the narrative surrounding this sterol.

Cholesterol is a membrane constituent widely found in biological systems which serves a unique purpose of modulating membrane fluidity, elasticity, and permeability. As such a large constituent of human organs and tissues, it should come as no shock that cholesterol has been widely reported and used as a carrier for drug-delivery. If you take a look at the molecular structure of cholesterol, you see that it is amphiphilic. The polar 3-hydroxy group and the non-polar hydrocarbon domain give cholesterol a unique set of properties making it particularly attractive in drug delivery mechanisms.

Cholesterol is highly biocompatible, stabilizes membranes, forms liposomes with increased rigidity, and promotes fusion processes making it a perfect component of lipid-based drug delivery systems. In addition to these properties, cholesterol has a high drug loading efficiency and can incorporate both hydrophilic and hydrophobic drugs. The surface of cholesterol-based carriers can also be functionalized to fine-tune circulation time and target tissues or cells. Due to these properties, cholesterol-based drug carriers have been designed as nanoparticles, micelles, and liposomal formulations for delivering antivirals, antimalarials, anticancer treatments, genetic material (DNA, mRNA, siRNA), and gene therapies.

Cholesterol can however present certain problems when used in human pharmaceuticals. Most cholesterol commercially available is derived from egg or wool grease (sheep derived). These animal sources are not ideal for human pharmaceuticals due to the potential viral contamination. Not to mention the increasing preference for non-animal derived products due to ethical or personal reasons. But where do you turn for a non-animal derived cholesterol?
