Vaccine research has been rapidly increasing since the early 1990s. Between 1993 and 1997, less than 20,000 papers and patents were published related to vaccines. Fast forward twenty years, between 2013 and 2017, nearly 90,000 papers and patents were published related to vaccines (Global Science and Technology Trends Report, CAS, n.d.). And with the recent COVID-19 pandemic and subsequent race to find a vaccine, these numbers are sure to soar even higher between 2018 and 2022. Avanti's primary role in the pandemic has been as a supplier of lipids for the lipid nanoparticle delivery system, but our lipid-based adjuvant systems are also being investigated for COVID-19 vaccines, as well as cancer treatments and rare virus vaccines. mRNA therapeutics are being investigated for various diseases such as cancer immunotherapy, protein replacement, and infectious diseases. One of the key benefits of using mRNA compared to non-viral DNA or adeno-associated virus (AAV) gene delivery systems is that mRNA does not have to gain access to the nucleus to undergo transcription. This usually means that protein production is more robust and much faster. mRNA therapy also has a lower risk of mutagenesis and thus has a safer genotoxic profile. Many of our previous articles have described the challenges associated with protecting mRNA in extracellular environments. Lipids have emerged as a highly attractive delivery vehicle for mRNA.
